CUSTOM SYNTHESIS PROTOCOL
Paradigm Organics is equipped and positioned to provide non-GMP custom synthesis services to clients. We offer services for custom synthesis of intermediates, metabolites, derivatives of lead compounds, reference standards and stable-isotope labeled compounds. Our custom synthesis platform consists of carefully laid out procedures, and rigorously implemented steps that ensure a robust system for providing consistent quality products and services to our clients. Whether for custom synthesis of intermediates, metabolites, derivatives of lead compounds, reference standards or stable-isotope labeled compounds, our approach is essentially the same.
A customer inquires from us for custom synthesis of a chemical compound. The client provides the structure of the compound with or without synthesis route.
REACTION DESIGN
Reaction design is implemented using databases and published literature including patent literature, as well as our know-how at Paradigm Organics. We have access to searchable reaction databases for easy search and retrieval of reaction information. In addition we have access to well-equipped libraries at three major universities that are within twenty-minute driving time from our facility. Reaction synthesis sequence is discussed within the core synthetic chemistry group and an optimal synthesis route is selected.
REACTION IMPLEMENTATION
Once the synthesis route is selected, the reaction is carried out in glass or stainless steel reaction vessels to produce 1-5 mg at the lower limit to ~2 kg at the upper limit.
SAMPLE PURIFICATION
Following reaction, purification is carried out in large glass columns for flash column chromatography as necessary. Distillations for liquids and re-crystallizations for solids are routinely used to purify the compounds that we synthesize. Depending on client requirements, compounds are further purified by HPLC. The purification method adopted is dictated by size of sample as well as client requirements.
SAMPLE ANALYSIS
Compounds synthesized are analyzed by NMR and MS. Samples are also analyzed by HPLC and HPLC/MS if specifically requested by client.
SAMPLE PACKAGING AND SHIPPING
Samples are packaged in glass vials or bottles. Each sample is shipped with a certificate of analysis, including NMR, LC/MS, Elemental analysis, etc, reports. Samples are shipped in heat-sealed plastic bags by overnight mail in most cases. The shipped material is accompanied with a shipping document giving all the data related to the shipped sample.
